Slovensko biokemijsko društvo organizira 111. društveno predavanje. Predavala bo dr. Katalin Monostory (Laboratory of Metabolic Drug-interactions, Research Center for Natural Sciences, Hungarian Academy of Sciences, Budimpešta, Madžarska). Predavanje z naslovom "Personalized medication using CYPtestTM -Diagnostic approaches of drug-metabolizing capacity for personalized medication of transplant patients" bo v predavalnici Inštituta za biofiziko Medicinske fakultete, Vrazov trg 2, Ljubljana, v ponedeljek, 7. 5. 2012 ob 12.00.
Povzetek predavanja:
Many undesired side-effects or therapeutic failures of drugs are the results of differences or changes in drug-metabolism, primarily depending on the levels and activities of cytochrome P450 (CYP) enzymes. The drug-metabolizing capacity of the liver highly influences drug-efficacy or toxicity particularly in the early post-operative period of patients underwent transplantation. The aim of our project was to predict therapeutic failures or severe adverse drug reactions by genotyping and phenotyping for CYP polymorphism resulting in reduced or no activity of the key drug-metabolizing enzymes. CYPtestTM the multistep diagnostic assay has been developed for the estimation of drug-metabolizing capacity of the liver. The complex evaluation of CYP-genotyping for frequent mutations in CYP genes and CYP-phenotyping (CYP mRNA expression) of leukocyte samples provides information about the current hepatic CYP-activities. Prospective investigation of CYP-status allows prediction of potential ‘poor metabolizer’ phenotypes and facilitates improvement of the individual recipient therapy especially in early post-operative period leading to the optimization of drug choice and/or dose for a more effective therapy, avoid serious adverse effects, and decrease medical costs. Tailored medication will eventually contribute to the improvement of graft and recipient survival.
